雌激素对奶山羊乳腺上皮细胞增殖及超微结构的影响

摘  要:
为了探讨不同浓度的17β-雌二醇对体外培养的奶山羊乳腺上皮细胞增殖和超微结构的影响,试验利用0,25,100,250,500μmol/L 17β-雌二醇处理奶山羊乳腺上皮细胞,采用MTT法检测细胞增殖情况,比色法检测细胞外乳酸脱氢酶(LDH)含量,并利用透射电子显微镜观察乳腺上皮细胞的超微结构。结果表明:雌激素对乳腺上皮细胞增殖有促进作用,而且细胞增殖率随着雌激素浓度的增加而增加。25,250,500μmol/L的17β-雌二醇对乳腺上皮细胞的损伤较小,而100μmol/L的17β-雌二醇会引起乳腺上皮细胞严重的病理组织形态性损伤。不同浓度的雌激素处理对乳腺上皮细胞的超微结构的影响也不同,250μmol/L和500μmol/L 17β-雌二醇处理的乳腺上皮细胞分裂加速,代谢增强,而且细胞核浆比例失调,部分细胞呈现瘤细胞;而100μmol/L的17β-雌二醇处理的乳腺上皮细胞代谢活动变慢,甚至出现细胞凋亡。说明一定浓度的雌激素不仅可以促进乳腺上皮细胞的增殖,增强细胞的代谢活动,而且对细胞的损伤较小。

摘  要:
To study the effects of different concentrations of 17β- estradiol( E2) on the proliferation and ultrastructure in the mammary epithelial cells of dairy goat,the mammary epithelial cells were treated with 17β- E2 at different concentrations including 0,25,100,250 and500 μmol / L,respectively. The proliferation of mammary epithelial cells was detected by MTT assay,and the content of extracellular lactate dehydrogenase( LDH) was detected by the colorimetric method,while the ultrastructure of the mammary epithelial cells was observed using transmission electron microscopy. The results showed the estrogen played a role in promoting the proliferation of mammary epithelial cells,and the cell proliferation rate was increased with the increase of the concentration of the estrogen. 17 β- E2 at concentrations of 25 μmol / L,250 μmol / L and 500 μmol / L could cause minor damage to the mammary epithelial cells,but 100 μmol / L of 17β- E2 could cause severe histopathological injury in the mammary epithelial cells. Different concentrations of the estrogen had different effects on the ultrastructure of mammary epithelial cells,the treatments with 17β- E2 at concentrations of 250 μmol / L and 500 μmol / L could accelerate the division of mammary epithelial cells and strengthen the metabolic activity,while the ratio of nucleus and cytoplasm was imbalanced,some cells exhibited tumor cells;the treatment with 100 μmol / L of 17β- E2 could slow down the metabolic activity of the mammary epithelial cells,this might cause cell apoptosis. The results indicate that a certain concentration of estrogen can promote the proliferation of the mammary epithelial cells and enhance metabolic activity of the cells,but has less damage on the cells.
