基因功能缺失技术及研究现状

摘  要:
基因功能缺失是目前研究基因功能最重要、最有效的手段之一。目前应用比较广泛的基因功能缺失技术主要有反义RNA技术、RNA干扰技术、ZFN技术、TALEN技术、CRISPR-Cas系统等。这些技术通过抑制或者沉默特定基因的表达,研究生物体相关表型变化,推测该基因的相关功能。在实际研究中,基因功能缺失往往和基因过量表达一起,为基因的功能研究提供最直接的证据,在生物、医学、农业等领域得到广泛应用。本研究系统介绍了几种常用的基因功能缺失技术的发展历程、技术原理及应用等,并展望未来的研究和应用。

摘  要:
Loss of gene function technology is one of the most important and efficient methods to study gene function at present.Now main widely used loss of gene function technologies are anti-sense RNA,RNA interference,ZFN,TALEN,CRISPRCas nucleases technology and so on.Those technologies aim to inhibiting or turning off the expression of the target gene,researching the change of the biological phenotype,and thus their related functions were speculated.In the practical studies,loss of gene function is always along with gene overexpression,providing the most direct evidence for gene function research,which was widely used in biology,medicine,agriculture and other fields.This paper systematically introduces the developing history,technological principles and application of some common loss of gene function technologies,and the protest of their future research and application is introduced.
