甘肃冬小麦品种(系)面筋强度、黄色素含量和PPO活性相关基因的分子检测

摘  要:
为了明确甘肃冬小麦品种(系)中品质相关基因的分布状况,提高品质育种效率,以141份小麦品种(系)为材料,利用高分子量麦谷蛋白亚基1Dx5的特异PCR标记、与黄色素含量相关的八氢番茄红素合酶(phytoene synthase,Psy)基因Psy-A1的标记YP7A及多酚氧化酶(polyphenol oxidase,PPO)活性基因Ppo-A1的标记PPO18分别进行1 Dx5、Psy-A1和Ppo-A1的基因型检测.结果表明:含1 Dx5基因的材料56份,分布频率为39.7%;Psy-A1位点存在Psy-A1a和Psy-A1b2种等位变异类型,分布频率分别为62.4%和37.6%;Ppo-A1位点存在Ppo-A1a和Ppo-A1b2种等位变异类型,分布频率分别为42.6%和57.4%.1 Dx5、Psy-A1和Ppo-A1的基因类型在不同地区分布频率存在明显差异,1 Dx5和Psy-A1b基因在天水地区频率最高,分别为51.0%和40.8%;Ppo-A1b基因在庆阳地区频率最高,为77.8%,Psy-A1a基因在3地区的分布频率明显高于Psy-A1b基因.参试材料中,聚合1 Dx5、Psy-A1b和Ppo-A1b基因的材料有19份,这些材料可作亲本资源,在小麦品种面筋强度和面粉色泽的改良中加以利用.

摘  要:
In order to define the distribution of genes related to quality traits and improve breeding efficiency,3gene specific markers for HMW-GS1Dx5,Psy-A1 and Ppo-A1 genes were used to detect the allelic variations at these loci in 141 Gansu wheat cultivars and advanced lines.The results indicated that the frequency of 1Dx5 gene was 39.7%,the frequencies of Psy-A1 agenotype with high yellow pigment content and Psy-A1 bgenotype with low yellow pigment content at Psy-A1 locus were 62.4% and 37.6%,respectively;the frequencies of Ppo-A1 agenotype with high PPO activity and Ppo-A1 bgenotype with low PPOactivity at Ppo-A1 locus were 42.6%and 57.4%,respectively.Significant differences among 1Dx5,Psy-A1 and Ppo-A1 were found in different areas,1Dx5 and Psy-A1 b were distributed mainly in Tianshui area,with frequency of 51.0% and 40.8%,respectively;Ppo-A1 bhad highest frequency of 77.8%in Qingyang area.The frequency of Psy-A1 awas obviously higher than that of Psy-A1 bin three areas.19 materials contented desired gene for 1Dx5 and alleles for Psy-A1 b and Ppo-A1 b,thus it is essential to combine the desired genes in a variety to improve the processing quality of Gansu winter wheat.The markers used in this study were all gene-specific,with good stability and high accuracy,and they can be efficiently applied in molecular marker-assisted breeding for wheat quality improvement.
