农杆菌介导的明恢63转化体系的建立

摘  要:
本试验优化了籼稻的农杆菌遗传转化系统,并以明恢63为材料进行了遗传转化。结果显示,从1100块愈伤组织中获得133块抗性愈伤,其抗性愈伤率为12.7/,分化率为81.3/,所获取的转基因植株后代的分离绝大多数符合孟德尔遗传规律。因此,此系统是一个较好的农杆菌介导的遗传转化系统,可用于基因功能验证、新基因在后代的遗传方式及共转化培养marker-free转基因植株。

摘  要:
In the study, Agrobacterium-mediated transformation of indica rice was optimized and Minghui 63 was transformed by this system. Results revealed that 133 resistant calluses were obtained from 1100 callus tissues. The transformation efficiency was 12.7% and differentiation efficiency was 81.3%. Besides, segregation ratio of progenies from single-copy transgenic plants fit for Mendal law of 3:1. This suggested that the transformation system was an efficient transformation system and could be used for gene function confirmation, genetic analysis of new genes, and production of marker-free transgenic plants by co-cultivation.
