虎雪兰与朱砂兰、蕙兰正反交育种及种子无菌萌发与增殖研究

摘  要:
[目的]研究虎雪兰与朱砂兰、蕙兰正反交育种及种子无菌萌发与增殖。[方法]以虎雪兰、朱砂兰、蕙兰‘绿春兰’为亲本,采用正反交杂交法进行育种试验得到果实,对成熟的种子进行无菌萌发试验,并对种子萌发获得的小苗进行增殖研究。[结果]朱砂兰为母本获得杂交种子无菌萌发最适培养基为:1/2MS+0.5 mg/L BA+0.2 mg/L NAA+3%花宝1号+0.05%碳粉;最佳增殖及生长培养基为:1/2MS+1.0 mg/L 6-BA+0.5 mg/L NAA+0.5 mg/L KT+3%花宝4号+0.05%碳粉。蕙兰‘绿春兰’为母本获得种子萌发最适培养基为:1/2MS+1.0 mg/L 6-BA+0.5 mg/L NAA+3%花宝1号+0.05%碳粉;最佳增殖及生长培养基为1/2MS+1.5 mg/L 6-BA+0.5 mg/L NAA+0.5 mg/L KT+3%花宝4号+0.05%碳粉。[结论]该方法为虎雪兰的种质资源栽培提供了理论依据。

摘  要:
[Objective] This paper aims to study the aseptic budding and fast propagation on Cymbidium tracyanum var huanghua × Cymbidium mastersii interspecific hybridization seed. [Method] Cymbidium tracyanum var huanghua,Cymbidium mastersii and Cymbidium faberi Rolfe‘Lv Chun Lan'were employed as parents. The hybrid seeds were tested using direct crossing and reciprocal crossing,then germinated and compared with different medium in axenic culture. [Result] The results show that the best medium for the seeds of Cymbidium mastersii in axenic culture is 1 /2MS + 0. 5 mg / L BA + 0. 2 mg / L NAA + 3% Hyponex 1 + 0. 05% C,the best medium for multiplication culture and differentiation is 1 /2MS + 1. 0 mg / L 6-BA + 0. 5 mg / L NAA + 0. 5 mg / L KT + 3% Hyponex 4 + 0. 05% C. The medium for the seeds of Cymbidium faberi Rolfe‘Lv Chun Lan'in axenic culture is 1 /2MS + 1. 0mg / L 6-BA + 0. 5 mg / L NAA + 3% Hyponex 1 + 0. 05% C,the best medium for multiplication culture and differentiation is 1 /2MS + 1. 5 mg / L 6-BA + 0. 5 mg / L NAA + 0. 5 mg / L KT + 3% Hyponex + 0. 05% C. [Conclusion] This method provides a theoretical basis for the germplasm resource cultivation of Cymbidium tracyanum var huanghua × Cymbidium mastersii.
