GM-CSF与生长抑素融合表达质粒对小鼠生长抑素抗体及IgG亚型的影响

摘  要:
将70只小鼠,随机分为7组,每组10只,雌雄各半。第1组为对照组,第2~4组分别肌肉注射pcS/2SS、pGM-CSF/SS和pGM-CSF加pcS/2SSDNA疫苗;第5~7组分别口服以减毒沙门氏菌为载体的pcS/2SS、pGM-CSF/SS和pGM-CSF加pcS/2SSDNA疫苗。结果表明:肌肉注射和口服pGM-CSF/SS和pGM-CSF加pcS/2SSDNA免疫小鼠均能有效激发免疫系统产生较强的免疫应答,与对照组相比,生长抑素抗体P/N值都有提高,且随着免疫时间的延长,呈现不断增高的趋势;同种疫苗以肌肉注射或口服途径免疫小鼠,血浆生长抑素抗体P/N值变化不显著;阳性鼠的比例以肌注pGM-CSF/SS为最高,达60%,pcS/2SS肌注组仅30%,其他各组分别为40%~60%,对照组没有出现阳性鼠;试验各组IgG1抗体的免疫反应高于IgG2a抗体,且IgG1的免疫反应,pcS/2SS和pGM-CSF/SS组高于pGM-CSF加pcS/2SS组;IgG2a的免疫反应,pGM-CSF/SS组高于pcS/2SS和pGM-CSF加pcS/2SS组。口服免疫组的IgG1和IgG2a免疫反应强于肌肉注射免疫组。说明基因佐剂GM-CSF可增强生长抑素基因疫苗的免疫反应,口服免疫pGM-CSF/SS、pGM-CSF加pcS/2SS可同时激发Th1型和Th2型免疫应答,GM-CSF和SS的融合表达质粒的免疫反应优于pGM-CSF和pcS/2SS的共免疫。

摘  要:
A total of 70 mice at the age of 22 days were divided randomly into 7 groups to explore the immuno-enhancement effect of genetic adjuvant GM-CSF on somatostatin(SS)DNA vaccine.All mice of 7 groups were given two immunizations at interval of 2 weeks.Animals in Group 1 as control were immunized with normal saline solution.Groups 2-4 were immunized with pcS/2SS(group 2),pGM-CSF/SS(group 3)and pGM-CSF+pcS/2SS(group 4)intramuscularly(i.m.),respectively,while groups 5-7 with pcS/2SS,pGM-CSF/SS and pGM-CSF+pcS/2SS delivered by attenuated Salmonella via oral administration,respectively.The results were as follows:Fusion expression plasmid of GM-CSF with SS and co-immunization of pGM-CSF with pcS/2SS significantly enhanced the P/N value of SS antibody by i.m.or oral administration as compared to the control group and immunization of pcS/2SS alone.Difference of the P/N value of SS antibody in mice plasma with the same kind DNA vaccine at different routes was not significant.Percent of mice with positive SS antibody of pGM-CSF/SS i.m.was higher than that of other groups(60%).Percent of pcS/2SS by i.m.and oral administration was 30% and 40%,respectively.None of the positive occurred in control group.IgG1 response was higher than IgG2a for all experimental groups.Responses of mice immunized with pcS/2SS and pGM-CSF/SS were higher than those of pGM-CSF/SS along with pcS/2SS for IgG1,while response of pGM-CSF/SS was higher than that of pcS/2SS and pGM-CSF along with pcS/2SS for IgG2a.IgG1 and IgG2a responses of oral administration were higher than those of intramuscular injection,indicating that immunization of pGM-CSF/SS and pGM-CSF along with pcS/2SS by oral administration can induce Th-1 type and Th-2 type response.In conclusions,genetic adjuvant GM-CSF can enhance the immune response of somatostatin DNA vaccine.The immuno-enhancement effect of oral administration is higher than that of the intramuscular injection.Immune response of fusion expression plasmid of GM-CSF and SS is superior to that of co-immunization of pGM-CSF and pcS/2SS.
