砂梨变异的SRAP初步鉴定及亲缘关系分析

摘  要：
为了鉴定砂梨变异类型在DNA水平上是否发生遗传变异,以砂梨的两个变异单株及25个梨代表品种为试材,通过正交试验设计优化了SRAP扩增体系,并从88对SRAP引物中筛选出22对适宜的引物进行了PCR扩增,找出差异性片段,并应用系统聚类法,分析其与其它梨品种的亲缘关系。结果表明,砂梨适宜的SRAP体系为25μL反应体积中Mg2+2.4 mmol·L-1、dNTPs 0.1 mmol·L-1、Taq酶1.5 U、模板DNA 80 ng、引物0.18μmol·L-1;与对照品种‘清香’相比,‘02-1’有7条差异性片段,‘02-2’有13条差异性片段,说明其均在DNA水平上存在差异;聚类分析表明,‘02-1’首先与‘清香’聚为一类,‘02-2’先与‘新世纪’和‘黄花’聚类,再与‘清香’聚类,因此,推断‘02-1’可能为‘清香’的营养系变异,‘02-2’可能为‘清香’的姊妹系。

摘  要：
To identify wether there exit genetic variation happened in Japanese pear(Pyrus pyrifolia Nakai),27 accessions including the two variants '02-1' and '02-2' and other twenty-five cultivars were studied by method of sequence-related amplified polymorphism(SRAP) markers.Firstly,the SRAP-PCR reaction system was opti-mized with orthogonal design.22 pairs of suitable primers screened out from 88 ones were used to detect the dif-ferential fragment among the pear cultivars.The genetic relationships between accessions were analyzed by hier-archical clustering.The result showed that the optimal SRAP-PCR system was composed of 2.4 mmol·L-1 Mg2+,0.1 mmol·L-1 dNTPs,1.5U DNA polymerase,80 ng DNA template,and 0.18 μmol·L-1 primer in 25 μL total reac-tion volume.Compared with pear cultivar 'Qingxiang',seven and thirteen differential fragments were detected in '02-1' and '02-2',respectively,which indicated that there was genetic variation at the DNA level.Variant '02-1' was closely related to 'Qingxiang';and variant '02-2','Shinseiki' and 'Huanghua' fell into a cluster firstly,and then clustered with 'Qingxiang'.It was postulated that '02-1' was a sport,and '02-2' might be a strain origmated from 'Qingxiang'.
