用甲酸水解制备重组牛乳铁蛋白肽LfcinB(17-41)

摘  要:
根据大肠埃希菌密码子偏嗜性合成了LfcinB(17-41)的基因序列,将该序列亚克隆到pGEX-4T-1载体中,在大肠埃希菌BL21菌株中用IPTG诱导表达。LfcinB与GST融合,两者之间加入了甲酸裂解位点。BL21在30℃下培养融合蛋白GST-LfcinB可溶性表达较好。纯化的GST-LfcinB用甲酸水解释放P1-LfcinB。抑菌试验显示P1-LfcinB对兔源致病性大肠埃希菌有抗菌活性。

摘  要:
According to the bias of codon utilization of Escherichia coli,a fragment encoding LfcinB(17-41)was synthesized,linked into vector pGEX-4T-1,transformed into E.coli BL21,expressed and induced by IPTG.A formic acid cleavage site was designed between the LfcinB and GST.Using formic acid treatment,P1-LfcinB was released and purified from the fused protein GST-LfcinB,the soluble expression production in E.coli BL21 under 30 ℃ cultural condition.Finally,P1-LfcinB showed some antibacterial activity against pathogenic E.coli from infected rabbits.
