新鱼腥草素钠对鸡外周血T淋巴细胞增殖的影响

摘  要：
采集健康鸡肝素抗凝血,按常规方法分离外周血淋巴细胞并于体外培养24 h后,加入不同剂量的新鱼腥草素钠/刀豆素A(ConA),继续培养44 h后,以MTT法测定新鱼腥草素钠对淋巴细胞增殖的影响。结果表明,新鱼腥草素钠在体外具有刺激淋巴细胞增殖的能力,且与ConA具有协同作用;进一步分析表明,不同给药剂量的新鱼腥草素钠对淋巴细胞的影响不同,其中以终浓度为250 mg/L效果最明显。

摘  要：
5 mL anticoagulant blood were sampled from the healthy chicken and the peripheral lymphocytes were collected by lymphocyte separation medium,then the cells were cultured for 24 h in RPMI-1640 cell culture medium at 39.5 ℃ with 5% CO2.The supernatant was remove and cells were resuspended the in equal volume complete medium and were added to 96 well culture plates with 80 μL/well,and different dose of sodium new houtluyfonate and/or concanavalin A(ConA) were mixed with the cells.Following incubation for 44 h,the lymphocyte proliferation was determined by MTT method.The results showed that sodium new houtluyfonate could induce lymphocyte proliferative response singly or cocultivated with ConA.In addition,the effects of different dose of sodium new houtluyfonate were dissimilar on improving lymphocyte proliferation,while the optimistic concentration is 250 mg/L.
