南海黄鸡肌抑素基因的克隆及序列分析

摘  要：
从南海黄鸡的鸡胚中抽提总RNA,采用RT-PCR技术成功扩增到肌抑素基因的cDNA序列(第76~1 128位碱基),将目的片断克隆到PMD18-T-Simple载体,转化大肠杆菌DH5α,筛选阳性克隆质粒并进行测序分析。结果表明,克隆的目的片断由1 053个核苷酸组成,与预期结果一致,经与GenBank中已注册的序列AF019621对比,同源性为99.6%,其相应氨基酸的同源性为99.4%。

摘  要：
Total RNA was extracted from the 14-day embryo of Nanhai Yellow Chicken.The fragment including No.76～1128 base of myostatin gene cDNA sequence was amplified by reverse transcription-polymerase chain reaction(RT-PCR),then was cloned into PMD18-T-Simple vector and was sequenced.The results showed that myostatin gene cDNA sequence cloned in the experiment consisted of 1 053 nucleotides and shared 99.6% homology with cDNA sequence of AF019621 in GenBank,the homology of myostatin amino acids squence between them was 99.4%.
