基于纳米金的显色基因芯片快速检测病原微生物

摘  要:
为构建一种新型的比色芯片,以实现对临床常见感染病原微生物的快速、准确地检测和鉴定。采用纳米金标记巯基修饰的各待检病原微生物的特异性基因片段,与相应的以各待检靶序列的特异性寡核苷酸探针构建成的基因芯片杂交,并通过结合银染反应将杂交信号被放大形成裸眼可见的显色信息来判读检测结果。该芯片技术检测时间短,最低检测值达100fmol/L。操作方法简单,不需要特殊设备,能部分满足临床检测的通量要求,具有很好的临床应用前景。

摘  要:
To develop a novel colorimetric DNA chip based on nanogold,which aimed to detect and identify pathogenic microorganism rapidly and exactly.The specific gene fragments modified by thiol of target pathogenic microorganism were labeled by nanogold,hybridized with specific oligonucleotide probes immobilized on DNA chip.Hybridization signal was amplified by the interaction between nanogold and silver stain reagent to form spot which could be detected with naked eyes.This gene chip has high sensibility and specificity and can be easily operated without special device.The sensitivity of this assay reached 100fmol/L.It can meet the flux demand of clinical detection and has good prospect of clinical application.
