摘  要:
矢车菊-3-芸香苷(C3Y)是桑椹花青素的主要成分之一。采用超声波辅助有机溶剂浸提法提取8个果桑品种桑椹鲜果中的C3Y,当料液质量浓度为0.125 g/mL时的提取效率较高。以体积比91∶9的0.2%磷酸-乙腈为高效液相色谱(HPLC)流动相可有效分离检测桑椹提取液中的C3Y。8个果桑品种桑椹鲜果中的C3Y含量存在明显差异,桑品种日本田橙的桑椹C3Y质量分数高达0.135 5%左右,而桑品种桂花蜜的桑椹C3Y质量分数仅有0.000 4%。常温保存桑椹鲜果中的C3Y不稳定,4 h后降解率达23.15%左右,48 h后降解率高达67.13%左右,所以采摘后的桑椹鲜果应及时提取C3Y,并在密闭、低温环境中储藏。体外试验结果表明,桑椹C3Y对羟自由基(·OH)具有明显的清除效果,但对超氧阴离子自由基(O2-)的清除作用不明显,对H2O2诱导损伤人胚肝细胞L-O2具有明显的促增殖保护作用,提示桑椹C3Y是桑椹花青素抗氧化及对H2O2诱导损伤肝细胞起保护作用的主要功能性成分。

摘  要:
Cyanidin-3-rutinoside(C3Y) is one of the main ingredients of mulberry anthocyanins. Ultrasound-assisted organic solvent method was applied to extract C3Y from mulberry fruit of 8 varieties. High extraction rate was achieved when material to solvent ratio was 0. 125 g /mL. C3Y extracted from mulberry fruit could be efficiently separated and determined by HPLC using 0. 2% phosphoric acid and acetonitrile(91∶ 9,V/V) as mobile phase. Significant difference was observed in C3Y content of mulberry fruit between different varieties,among which the highest content was 0. 1355% in Riben Tiancheng,and the lowest content was only 0. 000 4% in Guihuami. C3Y was not stable in mulberry fruit when preserved under room temperature after harvesting.The degradation rate was around 23. 15% after 4 h of preservation and up to 67. 13% after 48 h of preservation. Therefore,fresh mulberry fruit should be used to extract C3Y timely after harvesting. Moreover,the extracted C3Y should be stored under airtight and low temperature condition. In vitro experiment on C3Y activity indicated that mulberry fruit C3Y had an evident effect on scavenging hydroxyl radical(·OH),but had no obvious effecton scavenging superoxide anion radical(O2-). Mulberry fruit C3Y also showed a significant proliferative and protective effect against H2O2-induced injury on L-O2 human embryonic hepatic cells. These results suggest that mulberry fruit C3Y is the main functional ingredient of mulberry anthocyanins having antioxidative effect to H2O2and protective effect to injured hepatic cells.
