拟南芥耐硒突变体的鉴定及基因定位

摘  要:
在硒盐胁迫下通过根弯曲特性筛选获得拟南芥(Arabidopsis thaliana)耐硒突变体,并以突变体为母本与野生型(Ler)杂交获得基因型完全杂合的F1代;自交产生的F2代硒敏感,硒耐受的分离比为3∶1,表明该突变体是隐性单基因遗传。F2代选出132株具有耐硒特性的植株作为图位克隆的群体,从应用SSLP和In/Del分子标记进行基因定位,通过分析目的基因与各分子标记之间的连锁关系,发现耐硒突变体基因位于第I号染色体F12P19和NGA111的分子标记之间。

摘  要:
A selenate-torlerance arabidopsis mutant was obtained by using the root-bending trait under selenate stress. It was conformed that the selenate resistant characteristics could be steady inheritance. Then this mutant was crossed with the wildtype Ler. and F1 generation were created with heterozygous type completely. The F2 crossed by theirself were expressed the separation ratio of 3∶1 in response of sensitive selenium to resistent selenium,It were suggested that this mutant was a monogenic recessive mutation. Map-based cloning population were composed of the 132 selenate-tolerance seedlings in F2 generation. Using SSLP and In/Del based on PCR to preliminary chromosomal location. Through linkage analysis,it's indicated that this mutant gene was located on the chromosome I,which was mapped between molecular marker F12 P19 and NGA111.
