黄连素对β-羟丁酸诱导的牛巨噬细胞炎症反应的影响

摘  要:
为了研究黄连素对β-羟丁酸(β-HB)刺激牛巨噬细胞炎症反应的影响,试验先利用CCK-8法检测黄连素及β-HB处理牛巨噬细胞后对牛巨噬细胞活性的影响,然后采用不同浓度(5,7.5,10μmol/L)的黄连素预处理牛巨噬细胞1 h后,加入β-HB(2.4 mmol/L)刺激24 h,用CCK-8法检测细胞活性,用荧光定量法检测肿瘤坏死因子α(TNF-α)、白细胞介素6(IL-6)、白细胞介素1β(IL-1β)的mRNA表达水平,采用Western-blot方法检测NF-κB信号通路相关蛋白的表达。结果表明:黄连素(5,7.5,10μmol/L)及β-HB(2.4 mmol/L)不会对牛巨噬细胞的活性产生影响。黄连素能够抑制炎性细胞因子IL-1β、IL-6、TNF-αmRNA的表达水平,同时黄连素通过下调IκB和P65磷酸化水平,抑制NF-κB信号通路的激活。说明黄连素能够在体外抑制牛巨噬细胞的炎症反应。

摘  要:
In order to research the effect of berberine on inflammatory response of bovine macrophages stimulated by β-hydroxybutyrate(β-HB),CCK-8 method was used in this experiment to detect the viability of bovine macrophages treated by berberine or β-hydroxybutyrate. Then bovine macrophages were pretreated with different concentrations of berberine(5,7.5,10 μmol/L)for 1 h,and were followed by the stimulation of β-HB(2.4 mmol/L)for 24 h. Cell viability was detected by CCK-8 method and,the mRNA levels of tumor necrosis factor α(TNF-α),interleukin-6(IL-6)and interleukin-1β(IL-1β)were detected by fluorescence quantitative assay. Western-blot method was used to detect the related proteins in NF-κB signaling pathway. The results showed that there was no effect on the viability of bovine macrophage treated by berberine(5,7.5,10 μmol/L)or β-hydroxybutyrate(2.4 mmol/L). Berberine could inhibit the mRNA levels of IL-1β,IL-6 and TNF-α. Meanwhile berberine inhibited the activation of NF-κB signaling pathway by down-regulating the phosphorylation of IκB and P65. The results suggested that berberine can inhibit inflammatory response of bovine macrophages in vitro.
