甘肃棘豆生物碱对小鼠S_(180)肉瘤生长及p53蛋白表达的影响

摘  要:
【目的】探讨甘肃棘豆生物碱对小鼠S180肉瘤的作用及机制。【方法】将带有S180肉瘤的小鼠分为5组:生理盐水阴性对照组,甘肃棘豆生物碱低(0.25g/kg)、中(0.5g/kg)、高(1.0g/kg)剂量组及环磷酰胺阳性对照组,给药10d后,剖取瘤块、胸腺、脾脏、肝脏和肾脏,计算抑瘤率及胸腺、脾脏系数增长率和体质量增长率;采用HE染色对肿瘤、肝脏、肾脏进行病理观察;免疫组化SP法计p53阳性细胞数。【结果】以阴性对照组为基数,生物碱低、中、高各剂量组及环磷酰胺阳性对照组对肿瘤均有不同程度的抑制作用,抑瘤率分别为35.85%,45.28%,51.89%和66.04%(P<0.05或P<0.01);生物碱各剂量组和环磷酰胺组p53阳性细胞数也明显低于阴性对照组(P<0.05);胸腺和脾脏系数增长率及体质量增长率结果显示,甘肃棘豆生物碱对小鼠胸腺、脾脏和体质量增长无明显影响(P>0.05)。【结论】甘肃棘豆生物碱对小鼠S180肉瘤的生长具有一定抑制作用,其机制可能与抑制突变型p53蛋白的表达有关。

摘  要:
【Objective】The research was done to study the effect of alkaloid fromOxytropis kansuensis on mice bearing sarcoma 180 and its possible mechanism.【Method】The S180 bearing mice were divided into 5 groups.Ten days after drug adminstration,the tumor,thymus,spleen,liver and kidney were dissected,the inhibition rate of the tumor weight and the growing rate of the thymus coefficient,the spleen coefficient and the gaining in weight calculated the pathology sections of tumor of liver and kidney observed and the p53 positive cell number counted by immuno-chemical SP method.【Result】In comparison with control group,alkaloid and cyclophosphamide group showed an inhibition rate of 35.85%,45.28%,51.89% and 66.04%,respectively(P<0.05or P<0.01),and also showed a decrease of p53 positive cell numbers(P<0.05).The growing rate of thymus coefficient,spleen coefficient and the rate of weight gaining reflected the alkaloid fromO.kansuensis had not the obvious action to the thymus,spleen and the gaining in weight(P>0.05).【Conclusion】Alkaloid fromO.kansuensis showed an anti-tumor effect and its possible mechanism might be associated with the inhibition of the expression of mutated p53 protein.
