摘  要:
该研究旨在探究鲁中肉羊HIRA基因g.71833755 T>C和g.71874104 G>A两个位点多态性及其与产羔数之间的关系,以期为鲁中肉羊高繁殖力分子育种提供新的遗传标记。利用全基因组重测序结合Sequenom MassARRAY~? SNP技术对鲁中肉羊HIRA基因2个多态位点进行检测,并与其产羔数进行关联分析。结果表明:鲁中肉羊HIRA基因g.71833755 T>C位点存在TT、TC和CC三种基因型,g.71874104 G>A位点存在AA、AG和GG三种基因型。g.71833755 T>C位点在鲁中肉羊中表现为低度多态(PIC<0.25),该位点在鲁中肉羊中处于哈代温伯格不平衡状态(P<0.05);g.71874104G>A位点在鲁中肉羊中表现为中度多态(0.250.05)。HIRA基因g.71874104 G>A位点多态性与鲁中肉羊第2胎、第3胎产羔数显著关联(P<0.05),杂合型AG个体各胎产羔数均高于野生型GG和突变纯合型AA个体;g.71833755 T>C位点多态性与鲁中肉羊第2胎产羔数显著关联(P<0.05),杂合型TC个体第2胎产羔数均高于野生型TT(P>0.05)和突变纯合型CC个体(P <0.05)。综上,g.71833755 T>C位点的C等位基因和g.71874104 G>A位点的A等位基因可能是提高绵羊产羔数的潜在有效的DNA标记。

摘  要:
The aim of this study was to explore the polymorphisms of g.71833755 T>C and g.71874104 G>A loci in the HIRA gene of Luzhong Mutton sheep and their relationship with litter size,and provide new genetic markers for molecular breeding of high fecundity in Luzhong Mutton sheep.Sequenom Mass ARRAY~? SNP assay was applied to genotype g.71833755 T>C and g.71874104 G>A loci of HIRA gene. Then the association was analyzed between HIRA and litter size in Luzhong Mutton sheep.From the result of genotyping,three genotypes(TT,TC and CC) were found at g.71833755 T>C locus and three genotypes(AA,AG,GG) were found at g.71874104 G>A locus in Luzhong Mutton sheep. Population genetic analysis showed that g.71833755 T>C locus was at lowpolymorphism(PIC<0.25)in Luzhong Mutton sheep,but the g.71874104 G>A locus was at moderate polymorphism(0.25C locus of Luzhong Mutton sheep was under Hardy-Weinberg disequilibrium(P<0.05)and the g.71874104 G>A locus was under Hardy-Weinberg equilibrium(P >0.05).Association analysis indicated that the polymorphism of g.71874104 G>A was significantly correlated with the litter size of the second and third parity in Luzhong Mutton sheep(P<0.05),and the litter size of AG was higher than GG and AA for each parity;the polymorphism of g.71833755 T>C was significantly correlated with the litter size of the second parity in Luzhong Mutton sheep(P<0.05),and the litter size of TC was higher than TT and CC for each parity.In conclusion,the allele C at the g.71833755 T>C locus and the allele A at the g.71874104 G>A locus may be potentially effective DNA markers for increasing the litter size of Luzhong Mutton sheep.
