海带多糖对大鼠血管平滑肌细胞凋亡的影响

摘  要:
目的:研究海带多糖诱导血管平滑肌细胞凋亡及其调控机制。方法:在碱性成纤维细胞生长因子(bFGF)诱导血管平滑肌细胞增殖模型基础上,运用MTT法考察海带多糖对血管平滑肌细胞(VSMC)增殖的抑制作用;利用荧光倒置显微镜观察VSMC细胞形态学的变化;通过流式细胞术分析VSMC细胞周期的分布;采用Western blotting免疫印迹法探讨VSMC细胞凋亡相关蛋白Bax和Bcl-2的表达。结果:海带多糖抑制VSMC细胞增殖抑制。细胞形态学观察结果及流式、Western blotting免疫印迹分析结果表明海带多糖可诱导VSMC细胞凋亡。其凋亡机制与其诱导细胞周期阻滞于S期以及提高Bax/Bcl-2比率有关。结论:海带多糖可诱导VSMC细胞凋亡。

摘  要:
Objective: To explore the apoptotic mechanism induced by kelp(Laminaria japonica) polysaccharides in rat vascular smooth muscle cells(VSMC).Methods: The bFGF-induced proliferation of VSMC was evaluated by MTT assay.The morphological changes of VSMC were investigated using fluorescence microscope.The cell cycle distribution of VSMC was determined using flow cytometry.The apoptotic factors Bcl-2 and Bax were evaluated by Western blotting assay.Results: Kelp polysaccharides inhibited the growth of VSMC.Moreover,the results of cell morphology,flow cytometry and Western blotting assay indicated that kelp polysaccharides could induce VSMC apoptosis.The apoptotic mechanism was related to the induction of S phase arrest,as well as the increase of Bax/Bcl-2 ratio.Conclusion: Kelp polysaccharides can induce apoptosis.
