水溶性癞葡萄降血糖多肽MC2-1-5的分离纯化

摘  要:
以四氧嘧啶型糖尿病小鼠血糖浓度为筛选指标,通过超滤、凝胶色谱、半制备型反相高效液相色谱分离纯化得到癞葡萄中起降血糖作用的多肽MC 2-1-5。串联飞行时间质谱仪测得该降血糖多肽相对分子量为3 405.517 4 Da。动物试验结果表明,该肽在以2mg/kg剂量灌胃给予四氧嘧啶型糖尿病小鼠时,小鼠的血糖值在灌胃2和4 h后较相应的初始血糖值降低61.70%和69.18%。

摘  要:
With the blood glucose concentration of mice with alloxan-induced diabetes mellitus as screening index,the polypeptide MC2-1-5 with hypoglycemic effect in Momordica charantia L.Var.abbreviata Ser was separated and purified through ultrafiltration,gel filtration chromatography and semi preparative reversed phase liquid chromatography(RPLC).The relative molecular weight of the hypoglycemic polypeptide detected by series TOMFS was 3 405.517 4 Da.The results of animal experiments showed that when the mice with alloxan-induced diabetes mellitus was given the polypeptide dose of 2 mg/kg through stomach filling,their blood glucose values decreased 61.70% and 69.18% in 2 and 4 h after stomach filling than the corresponding initial blood glucose value.
