山银花组织培养继代增殖过程中内源激素含量变化的研究

摘  要:
以山银花良种‘金翠蕾’为试验材料,分析组培苗继代增殖过程中内源激素含量的变化。结果表明:内源ZR、ABA和GA_3在组培苗继代增殖分化丛芽过程中起主要调控作用,内源ZR、GA_3含量较高和内源ABA含量较低有利于其继代增殖分化丛芽,内源IAA含量高也有助于组培苗继代增殖分化和生长;内源IPA含量较高可促进组培苗启动培养,但与增殖分化丛芽的关系不紧密。在一个培养周期中,组培苗GA_3/ZR值主导其增殖分化丛芽,GA_3/ZR值较高有利于组培苗增殖分化和生长,增殖分化盛期时则需要GA_3/ZR值保持较低的水平;虽然GA_3/ABA值也呈现了相似的效应,但作用没有GA_3/ZR值明显。在继代增殖培养的前4个周期中,保持较高水平的GA_3/ABA值和IPA/ABA值能促进外植体启动培养和组培苗增殖分化丛芽。

摘  要:
The changes of endogenous hormone content in the process of proliferation in vitro culture were analyzed by using Lonicera macranthoides ‘Jincuilei' as the experimental material. The results showed endogenous ZR, ABA and GA_3 played a major regulatory role in the proliferation and differentiation of tissue culture seedlings. The higher content of endogenous ZR, GA_3 and the lower content of endogenous ABA were beneficial to the subculture and differentiation of buds. The high content of endogenous IAA also contributes to the proliferation, differentiation and growth of tissue culture seedlings. The higher endogenous IPA content was beneficial to the initiation culture of plantlets, but it was not closely related to proliferation and differentiation. The GA_3/ZR value predominated the proliferated and differentiated of buds in a culture cycle. The higher GA_3/ZR value was beneficial to the proliferation and differentiation, while the lower GA_3/ZR value was required during the peak period.The GA_3/ABA value also showed a similar effect to the GA_3/ZR value, but the effect was not so significant. In the first four cycles of subculture proliferation, maintaining high levels of GA_3/ABA and IPA/ABA can promote the initiation culture of explant and the proliferation and differentiation of plantlets.
