17α-甲基睾酮在猪毛发中蓄积规律的研究

摘  要:
为建立猪毛发中17α-甲基睾酮残留的液相色谱-串联质谱检测方法,研究多剂量内服、单剂量肌肉注射两种给药方式下,17α-甲基睾酮在猪毛发的蓄积规律及差异,选用体重为(20.0±3.5)kg的杜长大三元杂交猪6头,随机分为2组,一组按体重4 mg/kg内服17α-甲基睾酮、连续给药30 d,另一组按体重4mg/kg单剂量肌肉注射17α-甲基睾酮,毛发样品均采集到给药后第61天。样品处理后经液相色谱-串联质谱仪分析检测。以时间为横坐标、质量分数为纵坐标,分析17α-甲基睾酮在猪毛发中的残留规律。结果显示,17α-甲基睾酮在0.75~1 000 ng/g范围内线性关系良好(r2=0.999 1);检测限为0.30 ng/g,定量限为0.75 ng/g。多剂量内服给药组,第一次给药后第10天,毛发中药物质量分数达到峰值,而后出现下降,到约16 d后趋于稳定,停药后逐渐下降;单剂量肌肉注射给药组,药物质量分数在给药后第10天达到峰值,而后迅速下降;两种给药途径,在给药后第61天均仍能检测到17α-甲基睾酮。结果表明,建立的检测方法灵敏度较高;多剂量内服给药组在给药30 d及停药后31 d内、单剂量肌肉注射给药组在给药后61 d内均可在猪毛发中检测到17α-甲基睾酮,但不同给药方式之间蓄积差异较大。

摘  要:
To develop a liquid chromatography tandem mass spectrometer method for the determination of 17α-methyltestosterone in pig hair,and to observe difference of accumulation of 17α-methyltestosterone between different approaches,six pigs(20.0 kg±3.5 kg) were randomly assigned to two groups, three of which were fed with a mixture containing feed and 17α-methyltestosterone(4 mg/kg BW per day) for thirty consecutive days, and another three of which were given a single intramuscular injection with 17α-methyltestosterone. The pig hairs were collected over a period of 61 days. The hair samples were conducted and then analyzed by liquid chromatography tandem mass spectrometer system. The accumulation rule was analyzed by time as X-axis and concentration of 17α-methyltestosterone in pig hair as Y-axis. The correlation coefficient was 0.999 1 over a dynamic range of 0.75 to 1 000 ng/g; the limit of detection was 0.30 ng/g, and the limit of quantification was 0.75 ng/g. The peak of concentration appeared on day 10 and following a period of decline in the multiple dose oral 17α-methyltestosterone group, and then a leveling-off was observed on about day 16. The peak of concentration appeared on day 10 and then rapidly declined in the injection 17α-methyltestosterone group. 17α-methyltestosterone was still detected on day 61 in two groups. In conclusion, a higher sensitive method was obtained.17α-methyltestosterone was detected in pig hair during the treatment(30 days) and withdraw period(31days) in oral administration group and 61 days after single intramuscular administration. The significant diversity of the accumulation rule between different methods of administration was observed.
