铜在红耳龟体内的蓄积规律及对各组织器官的影响

摘  要:
为了研究铜(Cu)在红耳龟(Trachemys scripta elegans)体内的蓄积规律及对各组织器官的毒性作用,选取同一批次的32只年龄为2岁的红耳龟,体重范围为817~1 046 g,随机分成4组,即对照组(水中Cu浓度≤0.01 mg/L)和试验Ⅰ、Ⅱ、Ⅲ组(水中Cu浓度分别为1.0 mg/L、3.0 mg/L、6.0 mg/L)。试验后第150天取出各组织器官测定其Cu含量,并进行抗氧化功能的评价及病理学分析。结果显示,与对照组比较,各试验组红耳龟肝、肾、胃和小肠的Cu蓄积量呈浓度依赖性增加,且肝和小肠的Cu蓄积量显著增多(P<0.05),而心脏、肺、肌肉和胰腺的Cu蓄积量无明显变化。试验Ⅱ、Ⅲ组抗氧化功能下降显著(P<0.05),主要表现为T-AOC、SOD、GSH-Px活力显著降低(P<0.05),MDA含量显著升高(P<0.05),且具有明显的组织特异性,主要受影响组织是肝、肾、胰腺和小肠。其病理切片中表现损伤与抗氧化功能下降的组织结果一致,提示Cu引起红耳龟组织病变机制可能与抗氧化功能下降有关。

摘  要:
In order to study the bioaccumulation of copper(Cu)in different organs of Trachemys scripta elegans and toxic effects of Cu on the organs and tissues,32 two-years old turtles,body weight between 817 g to 1 046 g,were randomly divided into the control group(Cu concentration≤0.01 mg/L in water)and the experimental groupsⅠ,Ⅱ and Ⅲ(Cu concentration was 1.0 mg/L,3.0 mg/L and 6.0 mg/L in water respectively).On day 150 post-administration,organs were collected to measure accumulation of Cu and antioxidant function as well as histopathological changes.The accumulation of Cu in liver,kidney,stomach,and intestines increased with the increase of Cu concentration,but only the liver and the intestines have been significantly increased.And the accumulation of Cu in heart,lungs,muscles and pancreas remained the same.The antioxidative function of group Ⅱ and Ⅲ were significantly decreased,showing decrease of T-AOC,SOD and GSH-Px(P<0.05),but increase of MDA(P<0.05).Histopathological staining results showed that the groups Ⅱ and Ⅲ appeared pathological changes of tissues in liver,kidney,pancreas and intestines.The damaged tissues were consistent with the tissues that decrease of the antioxidative ability.It was suggested that the pathological mechanism of the lesion of the turtle caused by Cu could be related to the decreased antioxidant function.
