3种越橘(桔)茎尖初代培养与增殖研究

摘  要:
以红豆越橘、笃斯越桔和美登的茎尖为试材,对影响茎尖离体培养的取材时期、外植体的消毒、培养基和激素配比等因素进行研究的结果表明:休眠期水培促萌的嫩芽污染率较低,最利于茎尖诱导成活.适于灭菌的处理为:红豆越橘用75%酒精30 s+0.1%升汞2 min+次氯酸钠4 min;笃斯越桔和美登用75%酒精灭菌30 s+0.1%升汞灭菌处理4 min+次氯酸钠灭菌2 min.适于茎尖初代培养的培养基组合为:红豆越橘用WPM(改良)+6-BA 0.05 mg/L+IBA 0.5 mg/L+GA30.1 mg/L,诱导成苗率最高达84.42%;笃斯越桔、美登用WPM(改良)+6-BA 0.1 mg/L+IBA 0.5 mg/L+GA30.1 mg/L,成苗率分别为83.44%、92.76%.适合的增殖培养基组合为:红豆越橘、笃斯越桔用WPM(改良)+ZT 1.0 mg/L+GA3为0.1 mg/L,增殖系数分别为3.90、3.72,苗高分别为2.66、2.51 cm;美登用WPM(改良)+ZT 1.2 mg/L+GA30.2 mg/L,增殖系数为4.59,苗高达到2.74 cm.

摘  要:
Take Vaccinium vitis-idaea, Vaccinium uliginosum and stem tip as material, affecting the stem tip in vitro culture of materials, disinfection of explants, culture medium and hormone ratio were studied. The results showed that the dormant buds on the hydroponic promote of pollution rate is low, the most conducive to stem tip induced survival. Suitable for sterilization processing was: Vaccinium vitis-idaea with 75%alcohol 30 s+ 2 min+0.1% mercuric chloride sodium hypochlorite 4 min. Vaccinium uliginosum and climb 30 s+0.1% mercuric chloride with 75% alcohol sterilization sterilization 4 min + sodium hypochlorite sterilization 2 min. Suitable for the stem tip combination for original culture medium: Vaccinium vitis-idaea with WPM(improved)+6-BA0.05 mg/L+IBA+GA30.1 mg, 0.5 mg/L, inducing rate up to 84.42%; Vaccinium uliginosum, beauty's WPM(improved)+6-BA+IBA 0.5 mg, 0.1 mg/L+GA30.1 mg/L, rate of 83.44%, 92.76%respectively. Suitable for combination of multiplication culture medium for: red bean cranberry, Benedict bilberry with WPM(improved)+ZT 1.0 mg/L+GA3 was 0.1 mg/L, proliferation coefficient of 3.90 and 3.72,respectively seedling height 2.66 cm, 2.51 cm; Deng with WPM(improved)+ZT 1.2 mg/L+GA30.2 mg/L,proliferation coefficient is 4.59, seedling height is 2.74 cm.
