烟草青枯病拮抗真菌烟曲霉发酵条件的优化

摘  要:
烟曲霉(Aspergillus fumigatus)对烟草青枯病菌(Ralstonia solanacearum)有稳定的拮抗作用,在验证其发酵液对烟苗生长无害的前提下,利用四因素(发酵温度、发酵时间、接种量、装液量)三水平三重复正交试验,测定该真菌的最佳发酵条件。结果表明,烟曲霉的最佳发酵条件为:发酵温度28℃、发醇时间96h、接种量5%、装液量50mL(150mL容器),此时真菌粗提物产量达301.3μg,且抑菌活性最高,青枯病菌OD630平均值为0.370。试验还证明,装液量和发酵温度是影响烟曲霉发酵效果的主要因素。

摘  要:
One strain of fungus inhibiting Ralstonia solanacearum was previously selected among 59 strains,which were isolated from healthy soil collected in Chongqing tobacco-growing region.The rDNA ITS sequence indicated that the active fungus was Aspergillus fumigatus.In this study,the fermentation liquid of A.fumigatus was firstly proved to be harmless to tobacco seedlings,and then its fermentation conditions were optimized according to orthogonal experimental design for enhanced antimicrobial effect.The results suggested that the optimal cultivation conditions were temperature of 28℃,cultivation time of 96h,inoculation volume of 5% and medium volume of 50mL in 150mL fermentation flask.Under those conditions,the quality of crude extract was 310.3μg,and the crude extract had the highest inhibition activity,when the average OD630 of Ralstonia solanacearum was 0.370.The medium volume and culture temperature were the major factors that affected the productivity of A.fumigatus product.
