烟草4CL与虎杖STS融合基因的构建、表达及融合蛋白纯化

摘  要:
白藜芦醇具有抗菌、抗癌、抗肿瘤、保护心血管、抗氧化等众多药理作用。4-香豆酰辅酶A连接酶(4-Coumarate-CoA ligase,4CL)和芪合酶(Stilbene synthase,STS)是其生物合成的关键酶和限速酶。本研究利用悬挂PCR(Overlap PCR)技术将白藜芦醇生物合成的最后两个关键酶4CL和STS的基因融合,构建原核表达pET30a-Nt4CL::PcPKS5重组质粒,原核表达Nt4CL::PcPKS5融合酶蛋白。预测其共编码945个氨基酸,含15个氨基酸的过渡氨基酸链(Linker),编码蛋白质分子量(Mr)为102.8kDa,等电点(pI)为5.69。原核表达筛选结果表明:温度为25℃,IPTG浓度为1.0mmol/L,诱导起始OD600值在0.4~0.6之间,诱导4h时目标蛋白的表达量最高。细胞破碎后,经Ni 2+亲和柱纯化、PD-10柱脱盐后可纯化得到融合蛋白。本研究为通过分子生物学途径合成白藜芦醇奠定基础。

摘  要:
Resveratrol is a medicinal compound used for anti-bacterial,anti-cancer and cardiovascular protection.4-CoumarateCoA ligase(4CL)and stilbene synthase(STS)are the key and rate-limiting enzymes.In this paper,Nt4CL and PcPKS5were fused together by Overlap PCR,and a recombinant plasmid pET30a-Nt4CL ::PcPKS5was constructed.The fusion gene encoded 945amino acids,containing a 15amino acids linker.The fusion protein has a molecular weight(Mr)of 102.8kDa,and isoelectric point(pI)of 5.69.Prokaryotic expression results showed fusion protein reached the highest level under the condition of 25℃,1.0mM IPTG and 4hof induction.Fusion protein was purified with Ni 2+affinity column and desalted through PD-10column.Nt4CL ::PcPKS5fusion protein can be used in the biosynthesis of Resveratrol.
