鸡抗猪产肠毒素大肠埃希菌-沙门菌二联五价卵黄抗体的变化规律及喷干工艺的研究

摘  要:
为获得具有一定保护力的外源性抗体制剂,用于防治仔猪腹泻,利用产肠毒素大肠埃希菌(ETEC)标准菌株K88、K99、987P和导致仔猪副伤寒的2株沙门菌,制备产毒素大肠埃希菌-沙门菌二联五价的灭活苗,免疫接种待开产母鸡400羽,聚乙二醇沉淀法分离纯化卵黄抗体,然后采用直接试管凝集法检测不同时间的卵黄抗体(IgY)效价。将试验结束后无菌收集的全蛋液稀释过滤后,按照喷干设备进口温度165℃,出口温度85℃进行喷雾干燥,制备卵黄抗体粉,并对其进行表观性状及相关指标的测定。结果表明,免疫接种蛋鸡后,卵黄液中抗体IgY的效价呈现规律性变化,喷雾干燥获得的卵黄抗体粉表观性状较好,粗蛋白含量为12.6%,体外消化率高达95%,IgY的效价为1∶29。通过3次免疫接种获得了高免卵黄抗体,并进行了全蛋喷雾干燥工艺的摸索,制备出了效价较高的抗体制剂,为预防和治疗仔猪腹泻提供了一条途径。

摘  要:
In order to gain a exogenous antibody preparation with certain protection for controlling piglet diarrhea.400 hens were immunized by the combined vaccine prepared with ETEC K88,K99,987P and two strains of Salmonella which leading to piglet paratyphoid.The IgY antibody was extracted from the prolific eggs through PEG precipitation,and the titers of IgY at different periods were detected by tube agglutination test.The whole egg liquid which was collected under the condition of asepsis after the experiment filtration and dilution,and the spray-dry parameter was optimized.The temperature of inlet air was 165℃,and the temperature of outlet air was 85℃.The apparent shape and related indexes of yolk antibody powder were measured.The result showed that the titer of IgY appeared a regular variation.The apparent shape of yolk antibody powder was good.And the crude protein content was 12.6%,and in vitro digestibility reached 95%,and the titer was 1∶29.Highly immunized yolk antibody was obtained after immunization three times,and antibody products were prepared through detecting the technology of spay-drying which provides a solution to the current problems of piglet diarrhea.
