摘  要:
以乙醇为提取溶剂对荞麦壳黄酮的提取工艺进行优化,同时考察了黄酮得率与抗氧化活性的相关性。基于Plackett-Burman试验设计考察了各因素对荞麦壳黄酮得率、DPPH和ABTS自由基清除活性的影响。试验结果表明,提取次数是影响黄酮得率和抗氧化活性的显著因素(P<0.05),乙醇体积分数对3个响应值均无显著影响。最佳提取条件为乙醇体积分数30%,料液比(g∶m L)1∶20,提取时间10 min,提取3次,在此条件下荞麦壳的黄酮类化合物的得率为2.381%,且抗氧化活性较强。荞麦壳黄酮得率和抗氧化活性的相关性分析结果表明,荞麦壳对ABTS自由基及DPPH自由基的清除活性均与黄酮得率呈正相关,DPPH自由基的清除活性与黄酮得率相关度高,相关系数为0.703。

摘  要:
Extraction technology of flavonoids in buckwheat shell was optimized using ethanol as extraction solvent,and correlation between extraction rate and antioxidant activity of flavonoids was investigated. Plackett-Burman experimental design was based on effects of various factors on extraction rate,DPPH and ABTS radical scavenging activity of buckwheat shell. The results show that extraction times was a significant factor affecting extraction rate of flavonoids and antioxidant activity( P<0. 05),and effects of ethanol concentration on three response value were not significant. The optimum extraction conditions were 30% ethanol,ratio of solid to liquid( g ∶ m L) 1 ∶ 20,extraction for10 min,3 times,under which extraction rate of flavonoids of buckwheat shell was 2. 381% and antioxidant activity was strong. The results of correlation between extraction rate of flavonoids and antioxidant activity of buckwheat shell showed that ABTS and DPPH radical scavenging activities were positively correlated with content of extraction rate of flavonoids,the degree of correlation between DPPH free radical scavenging activity and extraction rate of flavonoids was high,and correlation coefficient was 0. 703.
