口疮病毒四川流行株的分离鉴定及其遗传进化分析

摘  要:
为了解四川地区口疮病毒(ORFV)流行毒株的遗传变异情况,从当地4个黑山羊养殖场采集疑似ORFV感染的羔羊唇部结痂组织,通过MDBK细胞分离培养、PCR、病毒效价测定、间接免疫荧光试验等方法进行分离鉴定,并对分离毒株的保护性抗原基因B2L、F1L进行克隆与序列分析.结果表明,成功分离到4株ORFV,并将其分别命名为SC-LZ1、SC-LZ2、SC-PZH1、SC-PZH2.序列分析结果显示,这4株分离毒株保护性抗原基因B2L、F1L相对保守,其B2L、F1L基因编码的氨基酸序列与参考株的同源性分别为96.3%~100%和94.2%~100%.本研究为四川地区羊口疮的防制奠定了基础.

摘  要:
In order to understand the genetic variation of the epidemic strains of orf virus(ORFV) in Sichuan Province,the scabs of kids suspected ORFV infection were collected from four local black goat farms.Orf viruses were isolated and identified by MDBK cells culture,PCR,viral titer measurement and indirect immunofluorescence,following which the protective antigen genes B2 L,F1 L of the isolates were cloned and sequenced.The results showed that four strains of ORFV were successfully isolated and named SC-LZ1,SC-LZ2,SC-PZH1 and SC-PZH2 respectively.The protective antigen genes B2 L and F1 L of four isolates were relatively conservative, shared amino acid sequences similarities of 96.3% to 100%and 94.2% to 100% with the reference strains, respectively.In conclusion, this study will facilitate the prevention and control of Orf in Sichuan Province.
