口疮病毒XC13株的分离鉴定

摘  要:
采用MDBK细胞对西昌市某山羊场疑似患有口疮的山羊病料进行分离培养,经动物回归试验、PCR、遗传进化分析等对分离毒株进行鉴定。结果表明,病料在MDBK细胞上产生致细胞病变;接种病料的山羊出现典型的传染性脓疱;PCR从分离毒株中扩增出F1L基因片段,该基因片段序列与口疮病毒(ORFV)参考株FJ-SL2、FJ-SL1的核苷酸序列的同源性分别为99.8%、99.5%,亲缘关系较近,遗传进化分析处于同一分支上。本研究从疑似口疮病料中成功分离鉴定出1株口疮病毒,遂将其命名为ORFV XC13。

摘  要:
The virus strain was isolated from crust materials with orf like-clinical symptoms from a goat farm in Xichang Prefecture of Sichuan Province and co-cultured with MDBK cells.And the virus isolates were identified using the animal regression experiment,PCR and phylogenetic analysis.In result,MDBK cell lines infected with the crust materials displayed cytopathic effects.The goat inoculated with the crust materials occurred to typical orf symptoms.The F1 Lgene was amplified by PCR from the isolate and shared a nucleotide identity of 99.8%and 99.5% with those of orf virus(ORFV)FJ-SL2 strain and FJ-SL1 strain respectively.The results of the phylogenetic tree analysis indicated that there was close genetic relationship between ORFV XC13 and FJ-SL2 strain,FJ-SL1 strain.It was concluded that the orf virus strain was successfully isolated and identified and named as ORFV XC13 strain.
