口疮病毒B4R锚蛋白抑制细胞NF-κB信号通路的研究

摘  要:
为明确口疮病毒(ORFV)编码的B4R蛋白调节细胞N F-κB活化的作用,以ORFV ORF008基因编码的B4R蛋白为研究对象,构建真核表达载体p CM V-Flag008,转染293T细胞,首先,利用双荧光素酶报告基因系统分析B4R蛋白对NF-κB信号活化的调节作用;其次,采用W estern-blot检测B4R蛋白对NF-κB信号通路中NF-κB p65蛋白核移位和IκBα蛋白磷酸化水平的影响。结果显示,B4R蛋白对NF-κB转录因子报告载体荧光素酶活性有明显的抑制作用(P<0.05);B4R蛋白能阻止IκBα蛋白磷酸化降解过程而抑制细胞NF-κB p65蛋白核移位。由此可见,ORFV编码的B4R蛋白能抑制细胞NF-κB信号通路的活化。

摘  要:
To illuminate the function of B4 R encoded by orf virus(ORFV)in regulating the activation of host cell NF-κB,ORF008 gene of ORFV was cloned into p CMV-tag2 B.Using a dual-luciferase reporter assay and western blotting to analyze the regulation of NF-κB activation and the nuclear translocation of p65 and the phosphorylation of IκBα in 293 T cells transfected with p CMV-Flag008,respectively.Results showed that the B4 R protein can significantly block the expression of NF-κB reporter luciferase activity(P<0.05),inhibit the degradation of phosphorylation IκBα and suppress the nuclear translocation of p65.In summary,B4 R protein can inhibit the activation of cell NF-κB signaling pathway.
