高压电晕电场对出芽短梗霉菌诱变的研究

摘  要:
[目的]通过研究高压电晕电场诱变对出芽短梗霉多糖产量的影响,筛选出高产优质的出芽短梗霉菌株。[方法]采用高压电晕电场对出芽短梗霉进行诱变,在临界放电电压下诱变不同时间,测定出芽短梗霉的性状和多糖产量的变化。[结果]在临界放电电压4.4 k V下的最佳诱变时间是60 min;经过2次连续诱变,多糖产量达到9.57 g/L,是原始菌株的3倍多,发酵液颜色为米白色,最终p H为4.2左右;连续传代培养10代,其生状和产量无明显变化,传代稳定。[结论]高压电晕电场对出芽短梗霉菌的普鲁兰产量提高有显著作用。

摘  要:
[Objective] The effects of corona treatment on pullulan production were studied to screen Aureobasidium pullulans with high yield and good quality. [Method] Using mutagenesis of Aureobasidium pullulans by corona,different time at the critical discharge voltage was processed,and the change of Aureobasidium pullulans traits and yield of polysaccharides were determined.[Result]The best mutation time in critical voltage 4. 4 k V was 60 min. After two consecutive mutagenesises,the polysaccharide production reached 9. 57 g / L,more than three times of the original strain. Fermentation liquor color was white,and the final p H was about 4. 2. After continuous passage culture of 10 generation,its shape and the yield had no obvious change,and the passage was stable. [Conclusion] Corona had a significant effect on the improvement of the yield of pullulan.
