复方抗菌药物药敏试验方法的建立

摘  要:
在WTO推荐的纸片扩散法(K-B法)的基础上,建立对不明成分和含量的抗菌药物复方制剂的药敏试验方法,为抗菌药物临床选择提供科学依据,并便于在临床兽医中推广应用。试验设4组(每组6个重复),分别以浓度为1/2.5a(a为产品使用说明指导的容水量(L/g)),1/5a,1/10a和1/20a(g/mL)的复方抗菌药物溶液制备药敏纸片。受试菌为2株禽致病性大肠杆菌。试验结果表明:1/5a组制备的药敏纸片(药物剂量(mg/kg)为药敏片含量的250倍),药敏结果更为稳定,不同药物间抑菌圈直径平均数差异也较大,可为兽医临床抗菌药物对比选择提供较好的量化依据;标示成分及含量相同的不同抗菌制剂(甚至同一厂家生产)的药敏试验结果差异极显著(P<0.01),说明主成分相同的不同抗菌制剂对同一株病菌的抗菌效力差异很大,也进一步证明用药敏试验结果指导临床选择用药的科学性和必要性。

摘  要:
Based on the disk-diffusion test(K-B method)recommended by WTO,the drug-susceptibility test on antibiotic compound preparations with unknown ingredients and quantities was established,which would provide a scientific basis for clinical selection of the preparations,and be easily popularized and applied to clinical veterinaries.Four groups(6 replicates per group)were set up,and antibiotic compound preparations were dissolved at the concentrations of 1/(2.5a)(a: the litres of water required to dissolve 1 gram of the antibiotics defined in product instruction),1/(5a),1/(10a) and 1/(20a)(g/mL) to prepare filter-paper disks for sensitivity test,respectively.Two strains of avian pathogenic Escherichia coli were used as testing bacteria.The results of drug susceptibility test demonstrated that 1/5a group(the drug dosage was 250 times that of each disk contained) was steadier.And the variation among the diameters of inhibitory zones from different preparations was greater,and therefore could provide a good quantitative basis for comparative selection of preparations in veterinary clinical practice.The difference of antimicrobial susceptibility test results among different preparations marked with the same ingredients and quantities(even from the same manufacturer) was very significant(P<0.01),indicating that the activity of different preparations with same primary ingredients against the same test strain was different greatly.It suggested that it was scientific and necessary to guide clinical selection of antibiotic compound preparations based on drug-susceptibility test results.
