富铁酵母发酵条件的优化

摘  要:
利用酵母菌具有较强富集微量元素的特性,试验对酵母菌富集微量元素铁的条件进行了优化。从7株酵母菌中筛选出耐铁能力和富集铁能力均较强的CRJ3作为试验菌株。结果表明,500 mL三角瓶中装入75 mL发酵培养基,pH为5.0,接种量为2%,在28~30℃,180 r/min条件下摇床培养,采用硫酸亚铁作为铁源,适宜浓度为800μg/mL。在此条件下,培养所得的酵母菌体生物量能达1.30 g/100 mL,铁含量能达35 mg/g左右,转化率达到56.0%。同时CRJ3菌株采用恒定pH发酵罐培养,酵母菌体生物量能达到1.45 g/100 mL,总铁含量能达500 mg/L左右,有机化程度约为98%。

摘  要:
To use the feature of yeast with has strong enrichment of trace elements,the experiment was conducted to optimize the conditions optimization of yeast iron-rich.The CRJ3 with strong iron-resistantand iron-rich ability from seven yeast as a pilot was selected.The cultural conditions were optimized as follows:cultural temperature 28 ~ 30 ℃,shaker culture 180 r/min,800 μg/mL FeSO4·7H2O,amount of medium 75 mL/500 mL flask,inoculums 2 %(V/V),initial pH 5.0.Under the optimized conditions,the biomass of iron-enriched yeast,chromium content and conversion were 1.30 g/100 mL,35mg/g and 56.0 %,respectively.The CRJ3 strains was cultured by using a constant pH value of fermentation in the fermentor.The biomass and iron content were 1.45 g/100 mL and 450 mg/L,organic iron was about 98 % of iron being organic.
