胶乳凝集试验诊断水牛巴贝斯虫病的研究

摘  要:
应用体外培养所得的可溶性牛巴贝斯虫(Brabesia bovis)抗原进行胶乳凝集试验的结果表明,本抗原与其它血液原虫,如伊氏锥虫(Trypanosoma evansi)、边缘无浆体(Anaplasma marginale)、双芽巴贝斯虫(B.bigemina)、环形泰勒虫(Theileria annulata)及瑟氏泰勒虫(T.sergenti)之间无交叉反应。非特异性凝集因子试验、胶乳凝集抑制试验也表明本抗原有较强的特异性。去脾水牛犊人工感染牛巴贝斯虫后第3天,应用胶乳凝集试验即可在血清中查到相应抗体,感染后12~15天抗体滴度达到高峰(1:2560);对成年水牛,感染后第8天应用本法即可查到抗体,并维持较高的抗体滴度达5个月。致敏抗原在4℃和20~30℃可分别保存6个月和3个月。此类抗原作为本病的诊断抗原是可行的。

摘  要:
A serological method—Latex agglutination test (LAT) has been established by ultilizing in vitro culture-derived soluble antigen of Babesis bovis for the diagnosis of babesiosis in buffalo. There is no cross-reaction between B. bovis and Trypanosoma evansi, Anaplasma marginale, B. bigemina, Thieleria annulata and T. sergenti, Three days after two splenectomized calves of buffalo were artificially infected, the antibody was detected by LAT, and the period of parasite emergence was synchronized with Period of antibody oecuring. The activity of the sensitized latex particle was found to be stable for 6 months at 4℃ or 3 months at 25~30℃. LAT was conducted with various types of serum. The conformation to positive serum was 100%. The detected results showed that the positive ratio was 85.2% in epidemic area and the negative ratio was 98.15% in babesiosis-free district. The results showed that in vitro culture-derived soluble antigen of Babesia bovis is effective as a diagnosis antigen.
