摘 要:
红松主产于黑龙江省中北部小兴安岭的珍贵树种。含有大量的酚类化合物,包括萜类、多酚类、生物碱等多种生物活性成分,具有抗辐射、抗氧化等多种生物学功能。本研究以小兴安岭的红松树皮为实验原料,采用乙醇溶剂萃取,并用大孔吸附树脂AB-8进行纯化,在此基础上用制备型高效液相法对红松树皮多酚进行二次纯化,分别考察流速、进样量、紫外线波长、洗脱浓度对色谱峰的影响,已确定最佳洗脱条件。结果表明最佳工艺条件为:流速为8mL/min,进样量为一针(0.25mL),紫外线波长为308nm,浓度洗脱为0~10min,0A/100B;10~40min,20A/80B;40~50min,60A/40B;50~60min,65A/35B;60~65min,95A/5B;65~100min,100A/0B。

摘 要:
Pinus koraiensis is a main rare species of north central part of Xiaohingganling,Heilongjiang.Pinus koraiensis contents a large amount of polyphenolic compounds,which are terpenes,polyphenols,alkaloids,etc.and these compounds possess a variety of biological functions.The present study aimed to extract polyphenols from the Pinus koraiensis grown in Xiaohingganling by using ethanol and then the so obtained samples,containing polyphenols,was purified in two continuous steps as follows: 1) macroporous resin AB-8 purification,and 2) semi-preparative high performance liquid chromatographic purification.The impacts of five factors,namely flow rate,injection volume,UV wavelength,and elute strength,on chromatographic peak were analyzed in order to determine the optimal elute condition.The results showed that the optimal elute condition are as follows: flow rate 8 mL/min,injection volume 0.25 mL,and UV wavelength 308 nm.The optimal gradient elution condition was 0-10 min,0A/100B;10-40min,20A/80B;40-50min,60A/40B;50-60min,65A/35B;60-65min,95A/5B;65-100min,100A/0B.
