几个籼稻良种胚乳蛋白高营养组分的生化研究

摘  要:
用IEF-PAGE与SDS-PAGE分析和制备分离、层析自动扫描、微量蛋白分离纯化、氨基酸自动分析、FAO/WHO评判蛋白营养价值标准及统计学方法,研究来源、类型不同的8个籼稻良种(着重其中4个)的胚乳蛋白不同等电点(pI)范围组分营养水平。结果表明:参试品种胚乳蛋白的等电点范围是pI8.6~4.0,含有15~17种等电聚焦组分;以弱酸性pI范围尤以pI6.0~5.2组分的必需氨基酸配比最佳—营养水平最高,其IEF组分含有16~26左右种多肽链;这个pI范围IEF组分的含量比率在参试品种之间及同一品种的两个种植年份之间较易变动,显示提高这个pI范围组分含量比率的可能性较大。初步发现,提高胚乳蛋白pI6.0~5.2组分的含量比率,对籼稻营养品质的常规的和现代生物工程的育种以及利用其他生物技术来提高稻米营养品质,可能具有重要理论与实际意义。

摘  要:
The analytic and preparative IEF-PAGE was used to separate the endosperm holo-protein of four excellent indica rice varieties one of them in two cropping years. The isoelectro-focusing spectrums showed that the endosperm proteins of these varieties (and other four accompanying experimental varieties) contained 15-17 focusing bands distributed at pI 8. 6-4. 0. The electrophoretical bands were scanned with a chromato automatic scanner, and were divided into five isoelectropoint ranges i. e. pI 8. 6-8. 0 pI8. 0-7. 2 pI6. 8-6. 0 pI6. 0-5.2 pI5. 2 -4. 0 after determination of the amounts of the bands. The amino acid species and contents of the protein bands in every pI range were separately determined with an automatic amino acid analyzer. The coordinate specific of essential amino acid was calculated according to the standard of FAO/WHO evaluation protein nutrient level. The results indicated that the focusing components at the weak acidic pI range in particular at pI6.0-5. 2 contained essential amino acids in optimal proportion (with the highest nutrient value), and the change of the content ratio of the protein components at this range of pI was easy between main experimental varieties (and one of them in two croppingyears). To raise the content ratio of the components at this pI range of the endosperm proteins has great significance.
