小鼠不同生长阶段骨骼肌组织中Myf5的表达

摘  要:
肌源性因子5(Myf5)是转录因子基本螺旋-环-螺旋家族的一员,它作为肌原性因子,对肌细胞的分化具有重要作用,在肌肉分化或肌细胞生成中也具有关键作用。试验分别选取幼年、性成熟、体成熟和老年期4个发育阶段的健康小鼠,采用HE染色、免疫组织化学、荧光定量PCR等方法主要研究Myf5在小鼠不同生长时期骨骼肌中的表达情况。结果显示,小鼠骨骼肌细胞核及肌纤维的生长在不同发育时期明显不同,其中,幼年小鼠的细胞核多位于细胞中央且排列较密,肌纤维的横切面近似圆形,性成熟、体成熟、老年的小鼠发育指标依次降低;Myf5主要在细胞核中表达,幼年期表达量高,老年期表达量最低;Myf5 m RNA在小鼠的不同发育阶段均有表达,但在不同阶段的表达量存在差异,其中,老年阶段的表达量最高,幼年、性成熟、体成熟阶段小鼠骨骼肌中Myf5表达水平极显著低于老年小鼠。Myf5的表达量与小鼠的生长发育阶段具有一定的相关性。

摘  要:
Myogenic factor 5 is a a key role in egulating muscle differentiation or myogenesis, specifically the development of skeletal muscle. And it is one member of the basic helix-loop-helix family of transcription factors, and it functions as a myogenic factor which is important for specification and differentiation of muscle cells. The aim of this experiment was to investigate the expression of Myogenic factor 5(Myf5)gene in mice skeletal muscle at different development stages by quantitative Real-time PCR, HE staining and immunohistochemistry. The results showed that the nuclei and muscle fibers of the skeletal muscle muscle of mice were significantly different at different development stages, among which the nuclei of the young mice were mostly located in the center of the cells and were closely arranged, the transverse plane of the muscle fibers was nearly circular, the sexual and physical maturation, and the development indexes of the old mice were decreased successively. Myf5 gene was mainly expressed in the early nucleus and the expression level was higher in childhood and the lowest in old age. Myf5 in mice were expressed in the different development stage, but there were difference in different stage of expressing quantity. The expression quantity of old age was the highest, Myf5 expression level in skeletal muscle of young, sexual maturation and body maturity was extremely significantly lower than that of old mice. The results indicated that the expression of Myf5 was correlated with the growth stage of mice.
