氨基酸对猪卵母细胞成熟及重构胚发育的影响

摘  要:
本研究旨在对比必需氨基酸和非必需氨基酸对猪卵母细胞体外成熟及重构胚发育的影响。在卵母细胞成熟培养液或重构胚培养液中分别添加1%的必需氨基酸或1%非必需氨基酸或同时添加2种氨基酸,无氨基酸组作为对照,分别统计卵母细胞成熟率和囊胚率。结果表明,在成熟培养液中添加非必需氨基酸组的卵母细胞成熟率为83.0%,显著高于其他各组(P<0.05);而在重构胚培养过程中,与对照组相比,非必需氨基酸组的囊胚率最高为20.3%(P<0.05),另外2组的囊胚率下降(P<0.05)。在卵母细胞成熟过程中添加2类氨基酸均能提高成熟率,但只有非必需氨基酸能够促进重构胚的发育。

摘  要:
The purpose of this study was to compare the effect of non-essential amino acids and essential amino acids on in vitro maturation and development of reconstructed embryos of porcine oocytes.The oocytes or reconstructed embryos were divided into four groups,then the IVM medium or embryo culture medium(NCSU-23)were added 1%the essential amino acids,1% non-essential amino acids,both 1% ssential and 1% nonessential amino acids,non-adding amino acids as control.After in vitro maturation of the oocytes or in vitroculture of the reconstructed embryos,the maturation rate and blastocysts rate were analysized.The results showed that the maturation rate of non-essential amino acid group was 83.0%,significantly higher than that of other groups(P<0.05);the blastocysts rate of the reconstructed embryonic was 20.3%in NCSU-23 with non-essential amino acids(P<0.05),while the blastocysts rate decreased in other two amino acids groups(P<0.05).The results indicated that both essential amino acids and non-essential amino acids can improve the rate of oocyte maturation,but only the non-essential amino acids can promote the development of reconstructed embryos.
