壳聚糖酶产生菌Mitsuaria sp.141-2的产酶发酵工艺

摘  要:
对产壳聚糖酶突变株Mitsuaria sp.141-2进行了研究,结果表明,该菌株所产的壳聚糖酶为诱导酶,最适作用pH值4.0,最适作用温度37~42℃。通过单因素试验和正交试验,得到优化的产酶条件:壳聚糖0.50/、装液量100mL·500mL-1三角瓶、起始pH值7.0、接种量2/、温度30℃、培养时间48h。在此条件下,酶活力可达3.631U·mL-1。

摘  要:
The mutant strain Mitsuaria sp. 141-2, with high yield of chitosanase, was researched and the result showed the chitosanase produced by the strain was an induced enzyme , the optimal pH and temperature were 7.0 and 37~42℃, respectively. By the single factor and orthogonal experiment, the optimal condition for the production of chitosanase was determined: colloid chitosan 0.50%, medium volume 100 mL in 500 mL flask, initial pH7.0, amount of inoculation 2%, temperature 30℃ and culture time 48 h. Under these conditions, the chitosanase activity reached 3.631 U·mL-1.
