培养基配方对冬虫夏草菌株产生粉红异变的影响研究

摘  要:
冬虫夏草真菌筛选过程中,蝙蝠蛾拟青霉SH-1的菌丝会逐渐产生粉红色变化,SH-1和粉红菌株ZH-1在形态上存在很大差异,经分子鉴定,两株菌为同一种。为查明产生粉红菌株的原因,最大化粉红菌株的得率,在考察4个培养基配方因素对粉红菌株产生的的单因素试验基础上,通过响应面法中的BBD设计优化冬虫夏草培养基配方,分析影响菌株SH-1产生粉色变化的因素。结果表明:产生ZH-1最适的培养基配方为氮源A(x_1)43.51 mL,K_2HPO_4(x_2)0.6 g、氮源B(x_3)2.56 g、葡萄糖(x_4)5.18 g。在此培养基配方条件下,菌株ZH-1产生概率为37.24%。试验结果表明,培养基配方中氮源A、K_2HPO_4的含量对产生粉红现象有显著影响,氮源A、葡萄糖含量对粉红菌株ZH-1的得率有显著影响。

摘  要:
A portion of fungi strain SH-1 would gradually change to pink color during the screening process of Cordyceps sinensis fungi.Although there was big morphological difference between strain SH-1 and pink strain ZH-1,molecular identification showed that the two strains were the same species.For seeking the reason of emerging pink variation and obtaining the maximum yield of the pink fungi,four main factors including nitrogen A,K_2HPO_4,nitrogen B and glucose were selected on the basis of experimental results of single factors through response surface methodology.The optimum culture conditions were as follow;43.51 mL nitrogen A,0.6 g K_2HPO_4,2.56 g nitrogen B,5.18 g glucose.Under the conditions,the optimum change probability of ZH-1 was37.24%.The results showed that the medium formula nitrogen A and K_2HPO_4 had very important effects on ZH-1 appearing probability,nitrogen A and glucose had very important effect on ZH-1 yield.
