猪A群轮状病毒LN-01-2013株的分离与鉴定

摘  要：
将病料与等体积2.5g/L胰酶溶液混合均匀,于37℃恒温培养箱中孵育1h,分别接种于MA-104、Marc-145、ST细胞系以分离病毒并筛选该毒株最适细胞系。通过RT-PCR以及重组质粒构建对其进行分子生物学鉴定以及遗传进化分析。结果显示,成功分离出猪A群轮状病毒LN-01-2013株,MA-104细胞系为其最适细胞系。LN-01-2013株的VP4基因型与P[7]基因型同源性最高,VP7基因型与G[9]基因型同源性最高。根据A群轮状病毒最新分类方法,LN-01-2013分离株属于P[7]G[9]型。

摘  要：
The equal amount of pathological sample and 2.5g/L trypsin solution were mixed uniformly at 37℃for one hour.Then the mixture was inoculated to MA-104,Marc-145and ST cells to isolate rotavirus and determine the best cell line,respectively.Molecular biology identification and analysis of genetic evolution were determined by RT-PCR and the constructed recombinant plasmid.A strain of group A porcine rotavirus named LN-01-2013was successfully isolated and the best cell line was MA-104.VP4gene of LN-01-2013strain showed the highest homology with P[7]genotype.VP7gene shared the highest homology with G[9]genotype.According to the recent classification of group A rotavirus,the LN-01-2013strain belonged to a P[7]G[9]genotype.
