甲氰菊酯降解酶的海藻酸钙凝胶固定化研究

摘  要:
海藻酸钙凝胶固定甲氰菊酯降解酶.实验室条件测定固定化酶对甲氰菊酯的降解特性.甲氰菊酯降解酶的固定化条件为:海藻酸钠质量浓度20 g/L;酶蛋白质量浓度1.0 g/L;固定化时间12 h;粒径5 mm;固定化酶对甲氰菊酯的降解pH稳定范围为5.0~10.0,稳定温度范围为25~50℃,适应性范围均较游离酶有所提高。Lineweaver-Burk法测定出固定化酶对甲氰菊酯的Km=298.13 nmol.mL-1,Vmax=43.48 nmol.mL-1.m in-1。最优条件下,固定化酶活力为游离酶活力的80%,制备的固定化酶可以重复连续利用3次。

摘  要:
Fenpropathrin degradating-enzyme was immobilized by Caalginate.And degradative characteristics of the immobilized enzyme were determined in the laboratory.Conditions for immobilization of fenpropathrin degradating-enzyme were with alginate:20 g·L~(-1),protein:1.0 g·L~(-1),time:16 h and diameter of enzyme particle 4 mm.Immobilized enzyme for fenpropathrin degradation was stable in pH range pH 5.0 to 10.0,temperature range 25℃ to 50℃,superior than dissociated enzyme.With Lineweaver-Burk method,immobilized enzyme had Km=298.13 nmol·ml~(-1),V_(max)=43.48 nmol·ml~(-1)·min~(-1).Immobilized enzyme vigor was 80% of dissociated enzyme in optimal conditions.Immobilized enzyme could be used 3 times continually.
