柚苷酶产生菌的抗药性UV诱变筛选

摘  要:
通过敏感抗生素对柚苷酶产生菌3-54菌株孢子致死浓度的测定,采用不同的UV照射时间对菌株孢子进行诱变处理,将诱变后的孢子涂在含纳他霉素致死浓度(2.0 U/mL)的平板筛选培养基上,获得了大量的抗药性致死突变株。从120个纳他霉素抗性基因突变株中通过透明圈初筛和多次摇瓶复筛,得到1株摇瓶发酵单位为500 U/mL的3-54-7号菌株,比出发菌株的产酶能力提高了近30%。

摘  要:
After testing the resistance of sensitive antibiotic to Naringinase-producing strains No.3-54,a lot of chemical resistant mutants were screened after the spores regenerated on the lethal media when they were treated with several different times of UV.A high-yield strain 3-54-7 was obtained from the 120 mutants,the productivity of which is 500 U/mL,almost 30% higher than that of the original strain in the rotation-flask experiment.
