响应面法优化巨大芽胞杆菌发酵培养基

摘  要:
为了提高微生物肥料生产菌株——巨大芽胞杆菌1013的发酵产芽孢数量,采用响应面法对其发酵培养基进行优化。首先采用单因子试验筛选出最佳碳源和氮源;然后设计基础培养基,采用Plackett-Burman方法对基础培养基中影响发酵产芽胞数各因素的效应进行评价,筛选出有显著效应的因素;最后通过响应面分析法优化主要因素的最佳浓度。结果表明,最佳碳源为糖蜜,最佳氮源豆饼粉和硝酸铵;基础培养基中对产芽孢数有显著效应的因素为糖蜜和豆饼粉,响应面优化后这2个因素的最佳含量为:糖蜜4.17%,豆饼粉1.53%。采用优化后的培养基进行摇瓶发酵,巨大芽胞杆菌的发酵产芽胞数为(89.34±3.87)×108cfu/mL,比基础培养基提高了3.35倍。

摘  要:
The response surface methodology was used to optimize the fermentation medium of Bacillus megaterium 1013. In the single factor experiment, molasses, soybean powder and NH 4 NO 3 , were selected to be the optimal carbon source and nitrogen source, respectively. Then, molasses and soybean powder were found to be the important factors by Plackett-Burman design. The optimal concentration of two important factors was determined by response surface methodology. The results showed that, 4.17% molasses, 1.53% soybean powder. The spores yield of Bacillus megaterium 1013 could reach (89.34 ± 3.87) × 10 8 cfu/mL by using the optimal medium, which was increased by 3.35 fold compared with the original medium.
