受MeJA诱导的烟草GTs基因启动子的克隆及转化

摘  要:
利用已克隆的1个受甲基茉莉酸(MeJA)诱导的糖基转移酶(GTs)基因,以PCR扩增得到该基因启动子序列,长度约为1.4kb,用改造后的双元载体pCAMBIA1301与GTs基因启动子连接,构建了含目的启动子与GUS基因的融合基因质粒pCAP-GUS。通过根癌农杆菌介导的烟草叶盘转化法转化烟草W38,经PCR检测验证得到了含有融合基因的转基因烟草植株,为进一步研究GTs基因的功能奠定了基础。

摘  要:
A plasmid pCAP-GUS was constructed by linking a 1.4Kb target promoter fragment,isolated from a methyl jasmonate(MeJA)-induced glycosyltransferase gene cloned by our lab,with the CaMV 35S promoter-deleted binary vector fragment in pCAMBIA1301.This recombined plasmid pCAP-GUS was then transformed into tobacco leaf discs of W38 mediated by Agrobacterium tumefaciens and the transgenic tobacco plants were achieved.It was suggested that the transgenic plants could be used as a model to study the function of MeJA-induced glycosyltransferase gene.
