超表达AtCpNifS基因烟草的淹水适应性

摘  要:
对超表达拟南芥半胱氨酸脱硫酶基因(Cysteine desulfurase gene of Arabidopsis,AtCpNifS)烟草(Nicotianatabacum Linn.)和野生型烟草进行了不同时间的淹水处理.结果表明,淹水处理的转基因和野生型烟草叶绿素质量比均减少,但转基因烟草的减少量小于野生型.淹水10 d,转基因烟草的根系活力有一定程度上升,淹水20 d则下降;淹水后野生型烟草根系活力一直呈下降状态;另一方面,淹水后乙醇脱氢酶(Alcohol dehydrogen-ase,ADH)和过氧化物酶(Peroxidase,POD)的活性均明显增加,转基因烟草的增加量较野生型烟草大.尤其是在淹水胁迫下转基因烟草表现的ADH酶活性明显增强的效应提示:超表达AtCpNifS基因对改善厌氧代谢可能有一定作用,说明转基因烟草表现出一定的淹水耐受适应性.

摘  要:
The transgenic tobacco(Nicotiana tabacum Linn.)overexpressing cysteine desulfurase gene of Arabidopsis(AtCpNifS) and the wild-type tobacco were treated by flooding with various time.The results showed that the chlorophyll content were reduced in both transgenic and wild-type tobacco.However,the reduction in transgenic plants were less than that in wild-type plants.The root vigor in transgenic tobacco showed a certain degree of rise after 10-d-flooding,and dropped after 20-d-flooding.The root vigor in wild.type tobacco gradually decreased after flooding.In contrast,the ADH and POD activities increased obviously after the flooding.Furthermore,the increments in transgenic plants were much more than that in wild-type tobacco.The ADH activity in transgenic tobacco was enhanced by flooding,suggesting that overexpression of AtCpNifS may be with some function of improving anaerobic metabolism,resulting in improved flooding tolerance in transgenic tobacco.
