α-酮戊二酸对脂多糖应激仔猪肠黏膜能量代谢的影响

摘  要:
试验研究日粮添加1%α-酮戊二酸(AKG)对多次脂多糖(LPS)刺激仔猪肠黏膜能量代谢的影响。选用24头体重(7.25±0.23)kg健康杜洛克×长白×大白断奶仔猪,随机分成3个组(对照组、LPS组、AKG组),每组8个重复,每个重复1头猪。对照组和LPS组饲喂基础日粮+1%淀粉,AKG组饲喂基础日粮+1%AKG,在试验第10天、第12天、第14天和第16天分别在LPS组和AKG组仔猪的腹膜注射80μg/kg LPS,对照组注射相应剂量的灭菌生理盐水。第17天仔猪麻醉后进行屠宰,刮取肠黏膜检测腺苷酸水平变化。试验结果显示,日粮中添加AKG可显著提高十二指肠黏膜二磷酸腺苷水平(P<0.05),并有效缓解LPS刺激导致的十二指肠和空肠黏膜的三磷酸腺苷和腺苷酸池水平的降低,提高空肠和回肠黏膜能荷水平。结果表明,仔猪日粮中添加1%AKG可以缓解LPS应激引起的肠黏膜能量代谢障碍,有利于肠黏膜屏障保护。

摘  要:
This experiment was conducted to study the effects ofα-ketoglutaric acid(AKG) on energy metabolism in intestinal mucosa of weaned pigs chronically challenged with lipopolysaccharide(LPS).Twenty-four healthy crossbred Duroc×Land-race×Largewhite weaned piglets with(7.25±0.23) kg BW were randomly allocated to 3 groups(control group,LPS group and AKG group).Each group included 8 replicates,1 piglet in per replicate.The control and LPS group were fed the basal diet supplemented with 1%starch,while the AKG group was fed the basal diet supplemented with 1%AKG.Piglets in the LPS group and AKG group were injected intraperitoneally with 80μg/kg LPS on day 10,12,14 and 16,whereas piglets in the control group were injected intraperitoneally with normal saline at the same dose.On day 17,the intestinal mucosa was collected after pigs were slaughtered under anesthesia.Then the mucosal samples were used to measure the levels of adenylate using HPLC method.The results showed that dietary supplementation of 1%AKG could significantly increase the levels of ADP in duodenal mucosa(P<0.05),significantly relieve the decline of the levels of ATP and total adenine nucleotide in duodenal mucosa and jejunal mucosa and improve the levels of energy charge in jejunal mucosa and ileal mucosa.The results indicated that dietary supplementation with 1%AKG could relieve the dysfunction of intestinal mucosal energy metabo- lism induced by LPS challenge,therefore exert protective effects on intestinal mucosal barrier.
