西藏绒山羊KAP7.1和KAP8.2基因多态性及其对部分生产性状的影响

摘  要:
为分析西藏绒山羊角蛋白关联蛋白(KAP)7.1基因和8.2基因与生产性状的关系,采用PCR-SSCP和DNA测序技术对340只西藏绒山羊KAP7.1基因和KAP8.2基因编码区序列进行多态性分析,并与体长、体高、胸围、产绒量、羊绒长度、羊毛长度等生产性状进行关联分析。结果显示:PCR-SSCP检测发现KAP7.1基因只有1种带型,KAP8.2基因有6种带型;KAP7.1基因扩增产物序列中未发现碱基变化;KAP8.2基因扩增产物序列中存在5个SNPs位点,分别为C115T、A207G、A214G、G216A和A278T,除了A278T外都位于编码区;对KAP8.2基因的基因型1、基因型2和基因型3与生产性能进行关联分析发现,基因型1个体的胸深和产绒量显著高于基因型3个体(P<0.05)。结果表明:KAP8.2基因可作为影响西藏绒山羊胸深和产绒量的分子辅助选择标记。

摘  要:
To analyze the relationship between keratin associated protein(KAP)7.1 and 8.2 genes and the production traits in Tibetan Cashmere goats, we use the PCR-SSCP and DNA sequencing technology to explore the polymorphisms of the KAP7.1 and KAP8.2 gene coded region in 340 Tibetan Cashmere goats, analyzing the relationships between the polymorphism of KAP8.2 and their body length, body height, chest circumference, cashmere yield, cashmere length and wool length. The PCR-SSCP detection revealed that KAP7.1 had only one type of band and KAP8.2 gene had 6 types. DNA sequencing results indicated that no change in base was found in the sequence of the products of KAP7.1 gene amplification. There were 5 SNPs sites in the sequence of KAP8.2 amplification, including C115 T, A207 G, A214 G, G216 A and A278 T. All of these SNP sites were located in the CDS region except A278 T. Analysis of the correlation between genotype 1, genotype 2 and genotype 3 of KAP8.2 and the production traits of the goats showed that the chest depth and cashmere yield of genotype 1 were significantly higher than those of genotype 3(P<0.05). These results suggested that the KAP8.2 gene might be used as a molecular assisted selection marker to regulate the chest depth and wool yield of Tibetan Cashmere goats.
