梅花鹿鹿茸蛋白聚糖的提取与分离

摘  要:
探讨梅花鹿(Cervus nippon)鲜鹿茸中蛋白聚糖的提取与分离方法,为深入研究和开发利用鹿茸提供了理论基础。采用Tris-HCl缓冲液匀浆提取梅花鹿鲜鹿茸,将粗提液上DEAE Sepharose FF离子交换柱,聚丙烯酰胺凝胶电泳检测分离组分。结果表明提取液中总蛋白聚糖含量为6.69 mg/g;DEAE Sepharose FF离子交换层析收集到三个蛋白聚糖组分Ⅰ、Ⅱ、Ⅲ,其蛋白聚糖含量分别占总蛋白聚糖含量的45.25%、25.15%、18.26%;聚丙烯酰胺凝胶电泳考马斯亮蓝和甲苯胺蓝染色均着色,蛋白聚糖Ⅰ核心蛋白分子量大约为40 kD,甲苯胺蓝染色显示3条单一宽带。DEAE Sepharose FF阴离子交换层析能用于梅花鹿鲜鹿茸中蛋白聚糖的分离,且此法快速简便。

摘  要:
To investigate the method of extraction and isolation of proteoglycans from velvet antlers of sika(Cervus nippon),so as to provide materials for deep research and exploiting of velvet antlers.Fresh velvet antlers were cut and stirred with 15 volumes of Tris-hcl buffer,then centrifugated.Proteoglycans were isolated by DEAE Sepharose FF chromatography and determined by SDS-polyacrylamide electrophoresis.Showed that the total proteoglycans were 6.69mg in 1g fresh velvet antler and they were isolated into fractionⅠ,Ⅱ,Ⅲ.Proteoglycans in each fraction was 45.25%,25.15%,18.26% of total proteoglycans respectively.Every fraction was stained with toluidine blue and each had a single broad band,fractionⅠwas stained with Coomassie blue and its core ptotein moleculer mass was about 40 kD but fraction Ⅱ,Ⅲ was barely stained.DEAE Sepharose FF chromatography can be used to isolate proteoglycans in velvet antlers,also it is an easy and fast way.
