一株低温脂肪酶产生菌的筛选鉴定及酶学特性

摘  要:
[目的]分离获得低温脂肪酶产生菌和研究其酶学特性。[方法]对采自阿勒泰等地区的75份土样中脂肪酶产生菌进行富集和筛选,目的菌株经16S rRNA基因序列比对鉴定后,对其产生的脂肪酶进行酶学特性分析。[结果]菌株N6-12归属于假单胞属菌,可能为新种;其产生的脂肪酶的最适催化温度为30℃;酶催化的最适pH为6.5;金属离子Cu~(2+)、Fe~(3+)等对酶有较大的抑制作用,而Na~+对酶有激活作用。[结论]菌株N6-12产生的脂肪酶属于低温脂肪酶。

摘  要:
[Objective]To isolate cold -adapted lipase -producing strain and define characterization of the lipase.[Method]The microorganisms produced lipase were enriched and isolated from 75 samples from Altay Prefecture in Xinjiang Uyghur Autonomous Region.According to 16S rRNA gene sequence analysis,one isolate was classified,and characterization of the lipase were measured.[Result]The results showed that N6 - 12 was identified as the Pseudomonas genus,and might be a novel species of the genus.The characterization of the lipase displayed that the optimum temperature and pH were 30℃and pH 6.5 respectively.The lipase activity was inhibited by some metal ion,such as Cu~(2+),Fe~(3+),and so on,whereas it was stimulated by Na~+.The enzyme was not stabile,and about 40%loss was observed when it was incubated at 30℃for 120 minutes.[Conclusion]The enzyme produced by N6 - 12 was cold - adapted lipase.
