巨大芽孢杆菌发酵产青霉素G酰化酶的条件优化

摘  要:
以牦牛乳酸水解酪蛋白为氮源,在单因素试验的基础上,采用响应面设计,对巨大芽孢杆菌产青霉素G酰化酶培养基及发酵条件进行了研究.结果表明:以葡萄糖4.28g/L、牦牛乳酸水解酪蛋白10g/L、苯乙酸18.43g/L、MgCl2·6H2O 0.5g/L、FeCl3·6H2O 0.002g/L为培养基,在三角瓶装液量19.3%、接种量5.6%、温度28℃、时间48h、摇床转速220r/min的条件下,所产青霉素G酰化酶活力为232.05U/L,与未添加牦牛乳酸水解酪蛋白对照组相比提高了42.25%.

摘  要:
Taking hydrolysed yak casein acid as the nitrogen source,on basis of single-factor test,using the response surface design,the medium composition and fermenting conditions of bacillus megaterium producing penicillin G acylase were studied.The results showed that,the penicillin G acylase activity 232.05U/Lwas obtained when medium composed of glucose 4.28g/L,hydrolysed yak casein acid 10g/L, phenylacetic acid 18.43g/L,MgCl2 ·6H2 O 0.5g/L,FeCl3 ·6H2 O 0.002g/L,under this condition of liquid volume 19.3%in eriochrome flask,inoculation 5.6%,fermentation at 28℃and 48has well as shaking speed 220r/min,compared with medium of no hydrolysed yak casein acid,the penicillin G acylase activity increased 42.25%.
